PlasmidFactory is the exclusive worldwide provider of the 2-Plasmid-System for production of AAV vectors and holds the exclusive license from the German Cancer Research Centre (DKFZ) for global production, distribution, and use of Helper & Packaging plasmids of the pDG and pDP family.
We offer a large variety of Helper & Packaging plasmids for the production of serotype-specific AAV vectors. These plasmids are available from our In-Stock service.
Advances in the field of gene therapy have demonstrated the potential of viral vectors to treat various monogenic, inherited diseases in human patients. Thanks to their unique characteristics, recombinant vectors derived from adeno-associated viruses (AAVs) represent a particularly effective technology for delivering therapeutic genes to their targets. Optimizing production of these recombinant AAV vectors to meet the demand for increasingly large quantities, while maintaining quality and cost-effectiveness, is a key challenge.
Our In-Stock service currently includes Helper & Packaging plasmids to produce serotypes 1-6, 8, and 9. We offer complete characterization of your DNA by LPS assay, BCA assay, UV spectroscopy, restriction analysis, and detection of RNA and bacterial chromosomal DNA by agarose gel electrophoresis.
These plasmids are also available on request in HQ (High Quality) grade for clinical applications.

AAV vector plasmids – keeping ITRs intact
PlasmidFactory’s proprietary ITRRESCUE® technology enables amplification of plasmid DNA containing sensi-tive inverted terminal repeats (ITR). While restriction digestion with SmaI is the standard quality control for ITR sequences, this method is not 100% accurate and does not allow quantification of the fraction of non-intact plasmids. PlasmidFactory’s self-developed Next Generation Sequencing (NGS) technology complements our fermentation process, allowing sequencing and quantification of ITR sequences while maintaining intact plasmids. GFP reporter gene cassettes with ITR2 flanked single strandet (ss) or self-complementary (sc) reporter genes are available In-Stock as minicircle or plasmid DNA.

Gene Delivery Vehicles – AAVs for long-term gene expression
Adeno-associated viruses have great potential in the field of gene therapy. The use of minicircles improves the quality of viral vector preparations and increases production efficiency.
When Minicircles were used for AAV production we got an increase of the genomic titer between 300-400% and an increase of the yield per cell.
